Postdoctoral Position Open in an NIH funded project in Dasgupta Lab: Understanding metabolic identity of normal mouse and human neural stem cells and brain cancer stem cells. The project is specifically geared towards examining the role of nutrient and energy sensors (e.g., AMP kinase and mTOR) in normal brain development and in the genesis of high grade brain cancer (glioblastoma). I am looking for a dedicated, energetic and ambitious person with some experience and aptitude in neuroscience mouse genetics. First hand experience in making knockout and knockin mouse models will be highly advantageous, but not necessary. Knowledge of basic molecular biology and biochemistry is necessary. Some knowledge in cell culture, flow cytometry, microscopy, and biochemical assays and cell signaling is desirable. Persons with their own fellowships may also apply. The Department of Surgery-Urology is accepting positions for post-doctoral fellows in the field of urologic oncology with an emphasis on renal and prostate cancers. The position will begin in July. We are seeking highly motivated candidates who will conduct basic research with a translational emphasis. Our laboratory interests include the metabolism of cancer cells. In particular, our laboratory is interested in the metabolic changes present in cancer cells and the mechanisms by which these changes occur. We are also interested in the role of putative oncometabolites- small molecules with potential transforming properties. Additionally, our laboratory is interested in mTOR signaling with an emphasis on the mTORC signaling complex 2 (mTORC2). Finally, our laboratory is also interested in the epigenetics of renal cancer as many histone modifying genes have recently been identified as mutated in renal cancer. Through gene complementation studies, we are interested in the role of chromatin biology in the pathogenesis of renal cancer.

Expertise with basic techniques utilizing DNA, RNA, protein, and tissue culture is required. Candidates need to demonstrate independence and the ability to communicate.

新規 NIH グラントの研究プロジェクト (Title: mRNA synthesis and capping in nonsegmented negative strand RNA viruses, PI: Tomoaki Ogino) を遂行するにあたり、ポスドク研究員を一名募集します。

[研究内容]　本研究室では、モノネガウイルスの RNA 依存 RNA ポリメラーゼ (L タンパク質) の構造と機能を解析しています。モノネガウイルス目には、狂犬病ウイルス、麻疹ウイルス、RS ウイルス、ニパウイルス、エボラウイルスなどのヒトに重篤な病原性を示すウイルスが多く含まれており、これらウイルスの L タンパク質は、ウイルスゲノム RNA・mRNA 合成および mRNA 修飾 (キャップ付加、キャップメチル化、ポリアデニル化) を担う多機能酵素であると考えられています。近年我々は、モノネガウイルスのプロトタイプであるウシ水疱性口内炎ウイルスを用いることにより、L タンパク質が宿主真核細胞とは全く異なる機構で mRNA キャップ付加反応を触媒することを明らかにしました (Ogino and Banerjee, 2007, Mol. Cell, 25, 85-97; Ogino et al., 2010, PNAS, 107, 3463-3468)。本プロジェクトでは、1) L タンパク質が触媒する個々の酵素反応の生化学的解析、2) L タンパク質の酵素ドメイン構造の解析、3) 変異 L タンパク質をもつ弱毒化��ウイルスの作成、4) L タンパク質のキャッピング酵素活性を標的にした抗ウイルス剤の探索などを行います。
